摘 要: 微生物和地球上所有生命体一样,与客观环境相互作用,构成一个动态平衡的统一整体,并在其中有一定规律性地分布、发育和参与各种物质循环。因此在一定的生态体系中,发育着不同特征性的微生物类群和数量,并在物质转化和能量转化中,呈现出各自不同的活动过程和活动强度。这种特征不仅受环境因子的直接或间接影响,而且由微生物本身所具有的适应性所决定。 关 键 词:微生物;持久性有机污染物;生物修复;净化作用
土壤中有机污染物 ,由于它的持久性和对人体 健康的潜在威胁 ,越来越受到人们的关注。残留在土壤中的有机污染物 ,不仅影响土壤的正常功能 ,降 低土壤环境质量 ,而且还可以通过生物富集进入食 物链 ,危及人体健康。因此 ,土壤有机污染物的去除 以及修复污染的土地 ,成为土壤环境研究领域的 一个重要研究课题。在污染土壤修复中 ,生物与物理化学修复相比具有非破坏性、经济性和安全性的 优点 ,使其成为有前途的污染环境修复技术之一。 土壤生物修复是利用土著性微生物、植物和小 动物等生物体 ,将土壤中的危险染物原位降解成 二氧化碳和水或转化成为无毒害物质 ,减少其对环 境风险的过程及其工程技术系统的影响。对于有机 污染土壤而言 ,生物修复就是利用土壤生物体将 机污染物做为唯一碳源和能源 ,或者通过共代谢作 用 ,从其它化合物获得碳源和能源后 ,降解有机污染 物大分子结构 ,使其成为简单、无害的形式[1 ] 本文在简单介绍微生物、植物修复和菌根修复 等三类生物修复方法特点的基础上 ,重点综述近期 微生物对土壤污染物修复的研究。
1 三类生物修复的概念
广义的生物修复,指一切以利用生物为主体的环境污染的治理技术。它包括利用植物、动物和微生物吸收、降解、转化土壤和水体中的污染物,使污染物的浓度降低到可接受的水平,或将有毒有害的污染物转化为无害的物质,也包括将污染物稳定化,以减少其向周边环境的扩散。一般分为植物修复、动物修复和微生物修复三种类型。根据生物修复的污染物种类,它可分为有机污染生物修复和重金属污染的生物修复和放射性物质的生物修复等。
狭义的生物修复,是指通过微生物的作用清除土壤和水体中的污染物,或是使污染物无害化的过程。它包括自然的和人为控制条件下的污染物降解或无害化过程。
植物修复:就是利用植物去治理水体、土壤和底泥等介质中的污染的技术。植物修复技术包括六种类型:植物萃取、植物稳定、根际修复、植物转化、根际过滤、植物挥发等技术。
微生物修复:即利用微生物将环境中的污染物降解或转化为其他无害物质的过程。
动物修复:指通过土壤动物群的直接(吸收、转化和分解)或间接作用(改善土壤理化性质,提高土壤肥力,促进植物和微生物的生长)而修复土壤污染的过程 污染物的污染水平[23 ] 。 所有这些研究都显示 ,直接分解土壤有机污染物是菌根真菌降解有机污染物的有效方式之一。降解的途径和机理涉及到菌根真菌的二次代谢产物和土壤酶反应 ,但对此了解甚少 ,需要更多的深入研究。
2.土壤中微生物的种类
土壤中微生物的类群、数量与分布,由于土壤质地发育母质、发育历史、肥力、季节、作物种植状况、土壤深度和层次等等不同而有很大差异。 l g 肥沃土壤,如菜园土中常可含有 10 8 个甚至更多的微生物,而在贫瘠土壤如生荒土中仅有 10 3 ~ 10 7 个微生物,甚至更低。土壤微生物中细菌最多,作用强度和影响最大,放线菌和真菌类次之,藻类和原生动物等数 量较少,影响也小。
它主要分为5类;(1) 、细菌 ;细菌积极参与着有机物的分解、腐殖质的合成和各种矿质元素的转化。( 2 )、放线菌 ;(3) 、真菌 ;(4) 、藻类 ;( 5 )、 原生动物

3.我国土壤污染的主要来源。
随着我国工业的发展。农业污染特别是土壤污染达到了惊人的水平。土壤污染的研究和治理已经受到了高度的重视,据出粗略统计,我国受重金属的污染的土地达到2000万公顷,受农药,化学试剂污染的农田达到了6000多万公顷。污染程度达到了世界之最,全国生产的农产品中有百分之20到60超标,我国农产品基本已无安全保障。每年大量的工业废水在我国环保措施还比落后的情况下随意的流入河流,再经过农民的灌溉流入农业用土,这又是增加我国土壤污染程度的重要原因。
现有的各种生物修复技术正是在这样的大环境下诞生的。但是由于各种原因无法得到大量的推广。本文主要阐述微生物修复技术的特点和前景为以后的发展方向出谋划策。
4.微生物的修复机制
污染环境的净化作用;环境自净是指环境受到污染后,在物理、化学和生物特别是微生物的作用下,污染物被逐步降解、消除并达到自然净化的过程。在环境自净中,微生物具有十分突出的作用。微生物的一大特点是其代谢类型多种多样。自然界中的各种物质,特别是有机化合物,几乎都可被微生物降解或转化。就是许多污染环境的人工合成物,也有微生物“正学着”如何分解。
土壤微生物的净化;天然土壤具有纯自然属性。人类最初开垦土地,主要是从中索取更多的生物量。在所开垦的土地逐渐变得贫瘠时,人们就向农田补充一些物质——肥料。在获得新肥力的同时,农田也受到了污染。譬如,施用人畜粪尿作肥料,可保持农田良好的生产性能,但病人的病原菌也可引起土壤的微生物污染。随着现代工农业生产的飞跃发展,施入农田的农药和化肥不断增加,土壤的污染程度日趋严重。目前,有杀虫效果的化合物已超过 6 万种,大量使用的农药也有 50 余种。农药对土壤的污染已引起土壤生产力和农产品质量的明显下降。 残留于土壤内的农药,经过生物主要是微生物的作用,经历种种复杂的转化、分解,最后将农药分解为二氧化碳和水。如果将土壤进行高压灭菌或采用抑菌剂处理,农药在土壤中的降解速度就会降低,甚至完全停止。研究表明,在未经消毒的土壤中,除草剂“敌草隆”的降解速度明显高于用氯化苦熏蒸消毒的土壤。前者, 6 周内敌草隆降解近半;而后者仅降解 1/10 。 微生物降解许多结构复杂的农药是借助共代谢作用进行的。所谓共代谢是指微生物在其他因子的协同作用下降解某些污染物的现象。其具体表现为:① 依靠环境提供营养物质。例如,只有在蛋白脂类物质存在时,直肠梭菌( Clostridium rectum )才能降解丙体 666 。 ② 依靠其他微生物协同作用。例如,链霉菌 ( Streptomyces ) 和节杆菌( Arthrobacter )可协作降解农药二嗪农的嘧啶环,两菌单独存在则均不能作用。③ 需有诱导物存在。例如,只有经正庚烷诱导后,铜绿假单胞菌( Pseudomonas aeruginosa )才能产生羟基化酶,使链烷羟基化为相应的醇。 微生物对有机氯农药 2,4-D 和有机磷农药对硫磷的降解,已有较多的研究报道。 2,4-D 是农业上广泛应用的具有高度选择性的内吸性除草剂。在高浓度下, 2,4-D 具有良好的除草效果,常用于杀除阔叶的双子叶植物;但低浓度 (1mg/L) 时则对植物有刺激生长作用,常用于促进早熟和生根,防治落花落果和倒伏等。 2,4-D 的微生物降解过程如图 11-2 所示。由于初期的微生物数量不多,降解十分缓慢,经过延缓期后,降解速度加快。至一个月左右, 2,4-D 完全消失。对硫磷是一类有机磷杀虫剂。微生物对这类杀虫剂降解明显快于有机氯农药。
污染环境的生物修复(bioremediation )早在上一世纪 80 年代就开始了。生物修复也曾称生物恢复 (biorestoration) 、生物清除 (bioelimination) 、生物再生 (bioeclamation) 和生物净化 (biopurification) 等。即是人为地利用和加强生物的代谢活动和其代谢产物降解和富集有毒有害污染物,从而恢复被污染环境的生产价值或景观价值的一个受控和自发进行的生物学过程。可利用于污染环境生物修复的生物可以有植物、动物和微生物。如利用芦苇发达的根系分解芳香族化合物,利用某些能富集重金属的植物来处理重金属污染土壤。利用蚯蚓分解农药污染土壤等。但微生物是污染环境自净和修复的主要贡献者。
污染环境生物修复可用原位 ( in situ ) 和异位( ex situ )或离位 ( off situ ) 两种不同方式进行。原位方式即是在污染环境原地进行技术性生物治理,不需将污染的土壤或水体转移。而异位方式是将污染土壤或水体转移至指定地点进行集中处理。生物修复的基本方法:一是进行生物扩增,即种植或接种具有降解和富集功能的植物或微生物;二是进行生物性刺激,即施加生物活性物质,刺激和促进土著微生物的生长和增殖,发挥其分解作用。
第一种方法是针对污染环境的主要污染物,选择具有降解这种污染物的微生物,通过发酵工程获得大量活性微生物,直接投加入污染土壤和水体,使污染环境中能降解这种污染物的微生物种群在数量上有极大的人为增强,促使污染物在较短时间内能得到有效降解乃至完全消除。第二种方法是运用给污染环境添加有针对性的营养物、电子受体和表面活性剂等物质,给污染环境中的有关微生物种群创造提高生物活性的条件,有利于微生物对污染物的降解和转化。在某些污染环境中由于污染物的不同,可能缺乏氮源物质,如原油污染环境;或者缺乏碳源物质和能源物质,如高氮施用环境。也有可能污染环境中缺乏微生物所需要的电子供体,如在厌氧环境中缺乏氧气,或 NO 3 - , SO 4 2- , CO 2 , Fe 3 + 等。许多污染物是非水溶性物质如石油、 PCBs 、 PABs 等,微生物难以接触污染物因而难以快速降解这些污染物。在生物修复过程中,如能有针对性的加入营养物或电子供体或表面活性剂,必将极大的有利于微生物的生长与降解污染物能力的提高。原位修复和异位修复都可通过这两种方法加速污染环境污染物的降解和消除。在大面积污染情况下,利用原位修复方法进行生物修复,如可用投加活菌,投加各种有效物质刺激微生物大量增殖,改善污染土壤和水体的通气条件,促进相关微生物的大量增殖与快速降解。在污染环境是一种少量可转移的情况时,可利用异位修复方式进行生物修复,如利用生物反应器法 (bioreactor) 、预制床法 (prepared bed) 、堆制法 (composting) 、生物堆层法 (biopiles) 等不同的方法。

Abstract : Due to the persistency of soil organic pollutants and their potential risk to the eco2environment and human health , there is an increasing urge to remediate organics polluted soils. Biodegradation of soil organic pollutants by rhizosphere microorganisms has been a research focus. Arbuscular mycorrhizal fungi are ubiquitous symbiotic microbes associated with plants. Compared with bacteria and actinomycete ,mycorrhizal fungi are more tolerant to organic pollutants in soil. For instance , some of them can use most persistent organic pollutants (POPs) as exclusive carbon source for obtaining energy. This paper reviewed the research into the relationships among mycorrhizal fungi , plant and organic pollutants during last two decades. The mechanisms of biodegradation of soil POPs by mycorrhizal fungi fall into three categories : (1) Direct degradation : mycorrhizal fungi can excrete enzymes to decompose organic pollutants to low molecular organic matter , carbohydrate , water and other non2toxic matter to obtain energy ; (2) Co - metabolism : mycorrhizal fungi use readily available or low molecular organics as primary carbon and energy source , and use high molecular organic pollutants as the second substrate ; (3) Degradation in the mycor - rhizosphere : the quantities of mi2 croorganisms in mycor - rhizosphere soil are up to 1000 times more than those in the bulk soils. Degradation of organic pollutants will be enhanced as a result of joint contribution from microbes living in the mycor - rhizo2 sphere. The inoculation of extraneous microbes such as nitrogen - fixing bacteria and genetically engineered bacteria can be used as a means of mycor2rhizosphere control to improve bioremediation efficiency. For instance , the in2 troduction of nitrogen2fixing bacteria will change the proposition of the bacteria in mycor2rhizosphere soil and re2 duce C/ N to improve the activities of soil microb
